The aim of the study was to determine the role of exosomes in modulating radiation resistance and their involvement in the radiation-induced bystander effect (RIBE) in PC3 and DU145 prostate cancer cells. The research focused on analyzing the impact of exosomes isolated from irradiated and non-irradiated cells on cell survival, apoptosis, and DNA damage response.
The study included clonogenic survival assays, apoptosis analysis by flow cytometry, and DNA damage assessment using γ-H2AX immunofluorescence. Exosomes were isolated via size exclusion chromatography (SEC) and characterized using nanoparticle tracking analysis (NTA) and immunoblotting. The results demonstrated that exosomes derived from irradiated cells induce a higher number of γ-H2AX foci, increase apoptosis, and affect recipient cell survival. It was also found that DU145 cells are more radioresistant than PC3 cells, and exosomes may contribute to increased radiation resistance in prostate cancer.
The dataset includes scanned Petri dishes with stained colonies, flow cytometry diagrams, NTA analysis data, as well as results from Western blot and γ-H2AX assays.
